How to prepare 1 M Tris-HCl pH 6.8
- Add the calculated Tris base to approximately 70–80% of the selected final volume of purified water.
- Stir until the Tris base dissolves completely.

- Bring the total solution volume to the selected final volume, mix thoroughly, and verify the final pH again.

Why is the HCl amount not calculated?
Tris-HCl refers here to a buffer prepared from Tris base and hydrochloric acid. The exact amount of HCl required depends on acid concentration, temperature, and pH adjustment conditions. A calibrated pH meter is needed to identify the endpoint accurately, but the calculator does not predict a titrant volume.
Is the 1 M pH 8.8 preset the standard Laemmli resolving-gel buffer?
No. This is an independently sourced 1 M Tris-HCl pH 8.8 formulation. Classical Laemmli resolving-gel stocks are more commonly prepared as 1.5 M Tris-HCl pH 8.8, so the two concentrations should remain separate presets.
Why should the temperature be noted when adjusting Tris pH?
